NM_002485.5(NBN):c.1284C>G (p.Asn428Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N428K variant (also known as c.1284C>G), located in coding exon 10 of the NBN gene, results from a C to G substitution at nucleotide position 1284. The asparagine at codon 428 is replaced by lysine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by BayesDel in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
