Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_000179.3(MSH6):c.1264G>A (p.Asp422Asn), citing Sema4 Curation Guidelines. This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 1264, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 422 with asparagine — a missense variant. Submitter rationale: To the best of our knowledge, the MSH6 c.1264G>A (p.D422N) variant has not been reported in individuals with MSH6-related disease. It was not observed in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 818734). In silico tools suggest the impact of the variant on protein function is deleterious, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
