NM_000051.4(ATM):c.124C>A (p.His42Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H42N variant (also known as c.124C>A), located in coding exon 2 of the ATM gene, results from a C to A substitution at nucleotide position 124. The histidine at codon 42 is replaced by asparagine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
