NM_177438.3(DICER1):c.115G>A (p.Asp39Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D39N variant (also known as c.115G>A), located in coding exon 1 of the DICER1 gene, results from a G to A substitution at nucleotide position 115. The aspartic acid at codon 39 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
