NM_000051.4(ATM):c.1078G>A (p.Asp360Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 1078, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 360 with asparagine — a missense variant. Submitter rationale: The p.D360N variant (also known as c.1078G>A), located in coding exon 8 of the ATM gene, results from a G to A substitution at nucleotide position 1078. The aspartic acid at codon 360 is replaced by asparagine, an amino acid with highly similar properties. In an assay testing ATM function, this variant showed a functionally normal result (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
