NM_015100.4(POGZ):c.375dup (p.Leu126fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.375dupA variant in the POGZ gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.375dupA variant causes a frameshift starting with codon Leucine 126, changes this amino acid to an Isoleucine residue, and creates a premature Stop codon at position 15 of the new reading frame, denoted p.Leu126IlefsX15. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.375dupA variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.375dupA as a pathogenic variant.
