Pathogenic — the classification assigned by GeneDx to NM_001356.5(DDX3X):c.373dup (p.Ser125fs), citing GeneDx Variant Classification (06012015): The c.373dupA variant in the DDX3X gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.373dupA variant causes a frameshift starting with codon Serine 125, changes this amino acid to a Lysine residue, and creates a premature Stop codon at position 5 of the new reading frame, denoted p.Ser125LysfsX5. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.373dupA variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.373dupA as a pathogenic variant.
