This variant lies in the ZIC1 gene (transcript NM_003412.4) at coding-DNA position 1184 through coding-DNA position 1194, deleting 11 bases; at the protein level this means shifts the reading frame starting at proline residue 395, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.1184_1194del11 variant in the ZIC1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.1184_1194del11 variant causes a frameshift starting with codon Proline 395, changes this amino acid to a Leucine residue, and creates a premature Stop codon at position 45 of the new reading frame, denoted p.Pro395LeufsX45. This variant is predicted to cause loss of normal protein function through protein truncation, as the last 53 amino acids are lost and replaced with 44 incorrect amino acids. The c.1184_1194del11 variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.1184_1194del11 as a pathogenic variant.
