Pathogenic — the classification assigned by GeneDx to NM_032436.4(CHAMP1):c.2079del (p.Ser694fs), citing GeneDx Variant Classification (06012015): The c.2079delC pathogenic variant in the CHAMP1 gene causes a frameshift starting with codon Serine 694, changes this amino acid to a Leucine residue and creates a premature Stop codon at position 9 of the new reading frame, denoted p.Ser694LeufsX9. This pathogenic variant is predicted to cause loss of normal protein function through protein truncation as the last 119 amino acids of the protein are lost and replaced with 8 incorrect amino acids. The c.2079delC variant is not observed in large population cohorts (Lek et al., 2016).
