Pathogenic — the classification assigned by GeneDx to NM_001127222.2(CACNA1A):c.4694dup (p.Pro1566fs), citing GeneDx Variant Classification (06012015): The c.4697dupC pathogenic variant in the CACNA1A gene causes a frameshift starting with codon Proline 1567, changes this amino acid to a Alanine residue and creates a premature Stop codon at position 29 of the new reading frame, denoted p.Pro1567AlafsX29. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.4697dupC variant is not observed in large population cohorts (Lek et al., 2016). Although this pathogenic variant has not been previously reported to our knowledge, its presence is consistent with the diagnosis of a CACNA1A-related disorder in this individual.
