This variant lies in the TRAPPC9 gene (transcript NM_031466.8) at 125 bases upstream of the translation start (5' untranslated region) through 124 bases upstream of the translation start (5' untranslated region), inserting TACCGTGAGGGTCACGGGGG. Submitter rationale: The c.170_171ins20 variant in the TRAPPC9 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant causes a frameshift starting with codon Tyrosine 58, changes this amino acid to a Threonine residue, and creates a premature Stop codon at position 29 of the new reading frame, denoted p.Tyr58ThrfsX29. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.170_171ins20 variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.170_171ins20 as a pathogenic variant.
