Pathogenic — the classification assigned by GeneDx to NM_001291415.2(KDM6A):c.2067_2074del (p.Asn690fs), citing GeneDx Variant Classification (06012015): The c.1911_1918delTAACTCCA variant in the KDM6A gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.1911_1918delTAACTCCA variant causes a frameshift starting with codon Asparagine 638, changes this amino acid to a Serine residue, and creates a premature Stop codon at position 15 of the new reading frame, denoted p.Asn638SerfsX15. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.1911_1918delTAACTCCA variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.1911_1918delTAACTCCA as a pathogenic variant.
