Pathogenic — the classification assigned by GeneDx to NM_001371928.1(AHDC1):c.1289del (p.Pro430fs), citing GeneDx Variant Classification (06012015): The c.1289delC pathogenic variant in the AHDC1 gene causes a frameshift starting with codon Proline 430, changes this amino acid to a Histidine residue and creates a premature Stop codon at position 22 of the new reading frame, denoted p.Pro430HisfsX22. This pathogenic variant is predicted to cause loss of normal protein function through protein truncation as the last 1174 amino acids are lost and replaced with 21 incorrect amino acids. The c.1289delC variant is not observed in large population cohorts (Lek et al., 2016).
