This variant lies in the SOX10 gene (transcript NM_006941.4) at coding-DNA position 1160 through coding-DNA position 1179, duplicating 20 bases; at the protein level this means shifts the reading frame starting at serine residue 394, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.1160_1179dup20 variant in the SOX10 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This frameshift variant replaces the typical last 73 amino acid residues in the SOX10 protein with 114 different amino acid residues. This alteration may interfere with the proper formation and/or function of the SOX10 protein. The c.1160_1179dup20 variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.1160_1179dup20 as a likely pathogenic variant.
