This variant lies in the ANKRD11 gene (transcript NM_013275.6) at coding-DNA position 6165, deleting one base; at the protein level this means shifts the reading frame starting at tyrosine residue 2056, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.6165delC variant in the ANKRD11 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.6165delC variant causes a frameshift starting with codon Tyrosine 2056, changes this amino acid to a Threonine residue, and creates a premature Stop codon at position 31 of the new reading frame, denoted p.Tyr2056ThrfsX31. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.6165delC variant is not observed in large population cohorts (Lek et al., 2016).
