Pathogenic — the classification assigned by GeneDx to NM_014795.4(ZEB2):c.2578_2579dup (p.Leu860_Asn861insTer), citing GeneDx Variant Classification (06012015). This variant lies in the ZEB2 gene (transcript NM_014795.4) at coding-DNA position 2578 through coding-DNA position 2579, duplicating 2 bases. Submitter rationale: The c.2578_2579dupCT pathogenic variant in the ZEB2 gene causes a frameshift starting with codon Asparagine 861 and changes this amino acid to a premature Stop codon, denoted p.Asn861Ter. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.2578_2579dupCT variant is not observed in large population cohorts (Lek et al., 2016).
