This variant lies in the NEB gene (transcript NM_001164508.2) at coding-DNA position 15781, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 5261 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The E5261X variant in the NEB gene has has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The E5261X variant is not observed in large population cohorts (Lek et al., 2016). We interpret E5261X as a pathogenic variant.
