NM_022893.4(BCL11A):c.314dup (p.Thr106fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.314dupT variant in the BCL11A gene has not been reported previously as a pathogenic variantnor as a benign variant, to our knowledge. The c.314dupT variant causes a frameshift starting withcodon Threonine 106, changes this amino acid to a Histidine residue, and creates a premature Stopcodon at position five of the new reading frame, denoted p.Thr106HisfsX5. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediatedmRNA decay. The c.314dupT variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.314dupT as a pathogenic variant.
