NM_014795.4(ZEB2):c.232del (p.Leu78fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.232delC variant in the ZEB2 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant causes a frameshift starting with codon Leucine 78, changes this amino acid to a Cysteine residue, and creates a premature Stop codon at position 11 of the new reading frame, denoted p.Leu78CysfsX11. The c.232delC variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. This variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.232delC as a pathogenic variant.
