Pathogenic — the classification assigned by GeneDx to NM_016628.5(WAC):c.313dup (p.Ser105fs), citing GeneDx Variant Classification (06012015): The c.313dupA variant in the WAC gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.313dupA variant causes a frameshift starting with codon 105 Serine, changes this amino acid to a Lysine residue, and creates a premature Stop codon at position five of the new reading frame, denoted p.Ser105LysfsX5. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.313dupA variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.313dupA as a pathogenic variant.
