NM_001368894.2(PAX6):c.609dup (p.Ile204fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.567dupC pathogenic variant in the PAX6 gene has been reported previously as an apparently de novo occurrence in association with aniridia (Dubey et al., 2015), and is consistent with the diagnosis in this patient. The duplication causes a frameshift starting with codon Isoleucine 190, changes this amino acid to a Histidine residue and creates a premature Stop codon at position 10 of the new reading frame, denoted p.Ile190HisfsX10. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.567dupC variant is not observed in large population cohorts (Lek et al., 2016). In summary, we consider this variant to be pathogenic.
