This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 2414, duplicating one base; at the protein level this means shifts the reading frame starting at arginine residue 806, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.2495dupG variant in the PLEC gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant causes a frameshift starting with codon Arginine 833, changes this amino acid to a Proline residue, and creates a premature Stop codon at position 11 of the new reading frame, denoted p.Arg833ProfsX11. The c.2495dupG variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. This variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.2495dupG as a pathogenic variant.
