This variant lies in the ASXL3 gene (transcript NM_030632.3) at coding-DNA position 1095 through coding-DNA position 1096, deleting 2 bases; at the protein level this means shifts the reading frame starting at glutamic acid residue 367, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.1095_1096delAA variant in the ASXL3 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant causes a frameshift starting with codon Glutamic Acid 367, changes this amino acid to a Glycine residue, and creates a premature Stop codon at position 17 of the new reading frame, denoted p.Glu367GlyfsX17. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.1095_1096delAA variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.1095_1096delAA as a pathogenic variant.
