Pathogenic — the classification assigned by GeneDx to NM_003482.4(KMT2D):c.16209_16222del (p.Ser5404fs), citing GeneDx Variant Classification (06012015): The c.16209_16222del14 pathogenic variant in the KMT2D gene causes a frameshift starting with codon Serine 5404, changes this amino acid to a Proline residue and creates a premature Stop codon at position 50 of the new reading frame, denoted p.Ser5404ProfsX50. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.16209_16222del14 variant is not observed in large population cohorts (Lek et al., 2016). Although this pathogenic variant has not been previously reported to our knowledge, its presence is consistent with the diagnosis of Kabuki syndrome in this individual.
