Submitter rationale: The c.2507_2508dupAA variant in the UBE3A gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.2507_2508dupAA variant causes a frameshift starting with codon Glutamic acid 837, changes this amino acid to a Lysine residue, and creates a premature Stop codon at position 5 of the new reading frame, denoted p.Glu837LysfsX5. This variant is predicted to cause loss of normal protein function through protein truncation, as the last 16 amino acids are lost and replaced with 4 incorrect amino acids. The c.2507_2508dupAA variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.2507_2508dupAA as a pathogenic variant.
