Pathogenic — the classification assigned by GeneDx to NM_015215.4(CAMTA1):c.4103_4104del (p.Glu1368fs), citing GeneDx Variant Classification (06012015): The c.4103_4104delAG variant in the CAMTA1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.4103_4104delAG variant causes a frameshift starting with codon Glutamic acid 1368, changes this amino acid to a Glycine residue, and creates a premature Stop codon at position 11 of the new reading frame, denoted p.Glu1368GlyfsX11. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.4103_4104delAG variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.4103_4104delAG as a pathogenic variant.
