The c.4255dupC variant is not observed in large population cohorts (Lek et al., 2016). The c.4255dupC likely pathogenic variant causes a frameshift starting with codon Histidine 1419, changes this amino acid to a Proline residue and creates a premature Stop codon at position 188 of the new reading frame, denoted p.His1419ProfsX188. This variant is predicted to cause loss of normal protein function, as the last 70 amino acids are replaced by 187 incorrect amino acids. Therefore, this variant is likely pathogenic; however, the possibility that it is benign cannot be excluded.
