Pathogenic — the classification assigned by GeneDx to NM_000094.4(COL7A1):c.6351delinsAT (p.Gly2117_Glu2118insTer), citing GeneDx Variant Classification (06012015). This variant lies in the COL7A1 gene (transcript NM_000094.4) at coding-DNA position 6351, replacing the reference sequence with AT. Submitter rationale: The c.6351delGinsAT pathogenic variant in the COL7A1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The c.6351delGinsAT variant changes codon Glutamic acid 2118 to a stop codon, denoted p.Glu2118Ter. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.6351delGinsAT variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.6351delGinsAT as a pathogenic variant.
