Likely pathogenic — the classification assigned by GeneDx to NM_016343.4(CENPF):c.1081G>T (p.Glu361Ter), citing GeneDx Variant Classification (06012015): The E361X variant in the CENPF gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The E361X variant is not observed in large population cohorts (Lek et al., 2016). We interpret E361X as a likely pathogenic variant.
