Pathogenic — the classification assigned by GeneDx to NM_006662.3(SRCAP):c.8427T>A (p.Cys2809Ter), citing GeneDx Variant Classification (06012015). This variant lies in the SRCAP gene (transcript NM_006662.3) at coding-DNA position 8427, where T is replaced by A; at the protein level this means converts the codon for cysteine at residue 2809 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The C2809X variant in the SRCAP gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This nonsense variant in the C-terminus is predicted to result in protein truncation, as the last 422 amino acids are lost. The C2809X variant is not observed in large population cohorts (Lek et al., 2016). We interpret C2809X as a pathogenic variant.
