Pathogenic — the classification assigned by GeneDx to NM_001378687.1(ATP2C1):c.457C>T (p.Arg153Ter), citing GeneDx Variant Classification (06012015): The R153X nonsense variant in the ATP2C1 gene has been reported previously in association with Hailey-Hailey disease (Hu et al., 2000; Hamada et al., 2008). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The variant is not observed in large population cohorts (Lek et al., 2016). We interpret this variant as pathogenic.
