NM_001164508.2(NEB):c.7235A>G (p.Tyr2412Cys) was classified as Uncertain significance for Nemaline myopathy 2 by Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard, citing ACMG Guidelines, 2015: The heterozygous p.Tyr2412Cys variant in NEB was identified by our study in the compound heterozygous state, with a likely pathogenic variant, in one individual with nemaline myopathy. The presence of this variant in combination with a reported pathogenic variant and in an individual with nemaline myopathy increases the likelihood that the p.Tyr2412Cys variant is pathogenic. This variant was absent from large population studies. Computational prediction tools and conservation analyses suggest that this variant may affect the protein, though this information is not predictive enough to determine pathogenicity.
