Uncertain significance for Familial hypokalemia-hypomagnesemia — the classification assigned by Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard to NM_001126108.2(SLC12A3):c.2273T>G (p.Ile758Ser), citing ACMG Guidelines, 2015: The heterozygous p.Ile758Ser variant in SLC12A3 was identified by our study in the compound heterozygous state, with a likely pathogenic variant, in one individual with Gitelman syndrome. The presence of this variant in combination with a likely pathogenic variant and in an individual with Gitelman syndrome increases the likelihood that the p.Ile758Ser variant is pathogenic. This variant has been identified in 0.001792% (2/111584) of European (non-Finnish) chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP rs61730207). Computational prediction tools and conservation analyses do not provide strong support for or against an impact to the protein.
