NM_000317.3(PTS):c.272A>G (p.Lys91Arg) was classified as Pathogenic for 6-Pyruvoyl-tetrahydrobiopterin synthase deficiency by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the PTS gene (transcript NM_000317.3) at coding-DNA position 272, where A is replaced by G; at the protein level this means replaces lysine at residue 91 with arginine — a missense variant. Submitter rationale: Variant summary: PTS c.272A>G (p.Lys91Arg) results in a conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251436 control chromosomes (gnomAD). c.272A>G has been reported in the literature in multiple individuals affected with 6-Pyruvoyl-Tetrahydropterin Synthase Deficiency (e.g. Ye_2013). These data indicate that the variant is very likely to be associated with disease. The following publication has been ascertained in the context of this evaluation (PMID: 23138986). ClinVar contains an entry for this variant (Variation ID: 813419). Based on the evidence outlined above, the variant was classified as pathogenic.
