Loss-of-function variation in CHAT is an established mechanism of disease in congenital myasthenic syndrome type 6 (PMID: 21786365). This variant has not been previously reported or functionally characterized in the literature to our knowledge. The c.982del (p.Asp328IlefsTer6) variant is present in the latest version of the gnomAD population database at an allele frequency of 0.0002% (3/1613988), and is absent in the homozygous state, thus is presumed to be rare. Based on the available evidence, c.982del (p.Asp328IlefsTer6) is classified as Likely Pathogenic.
