At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:36674751;31176036;17523162). It is absent or not present at a significant frequency in gnomAD. In silico models predict that this variant is possibly or probably damaging. In conclusion, we classify ABCB4 p.Lys1075Met (c.3224A>T) as a likely pathogenic variant.
