NM_147686.4(TRAF3IP2):c.1286C>A (p.Thr429Asn) was classified as Pathogenic for Discoid lupus rash by Nemer Genomics and Translation Biomedicine Lab, American University of Beirut, citing ACMG Guidelines, 2015: We have unveiled the first genetic clue in Discoid Lupus Erythematosus as being caused by a biallelic mutation in the gene encoding TRAF3IP2. Our mutation (p.T438N) segregates with the phenotype in a Lebanese family with 4 affected siblings and was absent from 300 ethnically matched individuals as well as in the gnomAD database with a null genotype frequency in gnomAD. Functional and expression studies in cultured cells revealed mis-localization of the mutated protein and a significantly reduced physical interaction with the interleukin 17-A receptor (IL17RA).
