Pathogenic for Polycystic kidney disease, adult type — the classification assigned by ARUP Laboratories, Molecular Genetics and Genomics, ARUP Laboratories to NM_001009944.3(PKD1):c.5141del (p.Phe1714fs), citing ARUP Molecular Germline Variant Investigation Process: The PKD1 c.5141delT; p.Phe1714fs variant has been described in at least one individual with autosomal dominant polycystic kidney disease (ADPKD; see link to Mayo ADPKD database). It is absent from general population databases (Exome Variant Server and Genome Aggregation Database), indicating it is not a common polymorphism. This variant causes a frameshift by deleting a single nucleotide, so it is predicted to result in a truncated protein or mRNA subject to nonsense-mediated decay. Additionally, several downstream truncating variant have been described in individuals affected with ADPKD and are considered pathogenic (Audrezet 2012, Rossetti 2007). Based on available information, the p.Phe1714fs variant is considered pathogenic.
