Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000162.5(GCK):c.755G>T (p.Cys252Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine, which is neutral and slightly polar, with phenylalanine, which is neutral and non-polar, at codon 252 of the GCK protein (p.Cys252Phe). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with GCK-related conditions. ClinVar contains an entry for this variant (Variation ID: 810090). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant disrupts the p.Cys252 amino acid residue in GCK.
