Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001270508.2(TNFAIP3):c.1033T>C (p.Tyr345His), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TNFAIP3 gene (transcript NM_001270508.2) at coding-DNA position 1033, where T is replaced by C; at the protein level this means replaces tyrosine at residue 345 with histidine — a missense variant. Submitter rationale: This sequence change replaces tyrosine, which is neutral and polar, with histidine, which is basic and polar, at codon 345 of the TNFAIP3 protein (p.Tyr345His). This variant is present in population databases (rs371548901, gnomAD 0.008%). This variant has not been reported in the literature in individuals affected with TNFAIP3-related conditions. This missense change has been observed in at least one individual who was not affected with TNFAIP3-related conditions (internal data). ClinVar contains an entry for this variant (Variation ID: 809999). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on TNFAIP3 protein function. Experimental studies have shown that this missense change affects TNFAIP3 function (external communication). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
