Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_015141.4(GPD1L):c.505+2T>G, citing Ambry Variant Classification Scheme 2023: The c.505+2T>G intronic variant results from a T to G substitution two nucleotides after coding exon 4 in the GPD1L gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, loss of function of GPD1L has not been established as a mechanism of disease. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
