Uncertain significance for Severe combined immunodeficiency, autosomal recessive, T cell-negative, B cell-negative, NK cell-negative, due to adenosine deaminase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000022.4(ADA):c.992A>C (p.Lys331Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine with threonine at codon 331 of the ADA protein (p.Lys331Thr). The lysine residue is moderately conserved and there is a moderate physicochemical difference between lysine and threonine. This variant is present in population databases (rs372131830, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with ADA-related conditions. ClinVar contains an entry for this variant (Variation ID: 809251). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
