Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_015311.3(OBSL1):c.3402del (p.Thr1135fs), citing LabCorp Variant Classification Summary - May 2015: Variant summary: OBSL1 c.3402delC (p.Thr1135ProfsX4) results in a premature termination codon, predicted to cause a truncation of the encoded protein, however the significance of this region is unclear. The variant allele was found at a frequency of 1.6e-05 in 242482 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.3402delC in individuals affected with Three M Syndrome 2 and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 809173). Based on the evidence outlined above, the variant was classified as uncertain significance.
