NM_000393.5(COL5A2):c.3200G>A (p.Arg1067His) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1067H variant (also known as c.3200G>A), located in coding exon 45 of the COL5A2 gene, results from a G to A substitution at nucleotide position 3200. The arginine at codon 1067 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
