Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014975.3(MAST1):c.1565G>A (p.Gly522Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 522 of the MAST1 protein (p.Gly522Glu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with MAST1-related conditions (PMID: 32198973). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 808458). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
