NM_001163435.3(TBCK):c.1880T>G (p.Phe627Cys) was classified as Uncertain Significance for Hypotonia, infantile, with psychomotor retardation and characteristic facies 3 by Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard, citing ACMG Guidelines, 2015: The p.Phe627Cys variant in TBCK has not been previously reported in the literature in individuals with TBCK-related intellectual disability syndrome, but has been identified in 0.0003% (4/1172420) of European (non-Finnish) chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP rs1579198426). Although this variant has been seen in the general population in a heterozygous state, its frequency is low enough to be consistent with a recessive carrier frequency. This variant has also been reported in ClinVar (Variation ID: 807702) and has been interpreted as pathogenic by Institute of Human Genetics (Klinikum rechts der Isar). Computational prediction tools and conservation analyses suggest that this variant may impact the protein, though this information is not predictive enough to determine pathogenicity.
