Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001354768.3(NRL):c.375C>G (p.His125Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine, which is basic and polar, with glutamine, which is neutral and polar, at codon 125 of the NRL protein (p.His125Gln). This variant is present in population databases (rs201970559, gnomAD 0.2%). This missense change has been observed in individual(s) with autosomal dominant retinal disease (PMID: 15591106). ClinVar contains an entry for this variant (Variation ID: 807100). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Experimental studies have shown that this missense change affects NRL function (PMID: 17335001). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
