This variant lies in the CYP11B1 gene (transcript NM_000497.4) at coding-DNA position 476, where C is replaced by T; at the protein level this means replaces proline at residue 159 with leucine — a missense variant. Submitter rationale: Variant summary: CYP11B1 c.476C>T (p.Pro159Leu) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251232 control chromosomes. c.476C>T has been reported in the literature in a homozygous individual affected with non-classical Congenital Adrenal Hyperplasia (Parajes_2010). These data indicate that the variant is likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in 10%-<30% of normal 11OHD activity (Parajes_2010).
