NM_004006.3(DMD):c.10498_10499del (p.Glu3499_Ser3500insTer) was classified as Pathogenic for Duchenne muscular dystrophy by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Ser3500*) in the DMD gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in DMD are known to be pathogenic (PMID: 16770791, 25007885). This variant is not present in population databases (ExAC no frequency). This premature translational stop signal has been observed in individual(s) with dystrophinopathies (PMID: 17041906, 18430534). In at least one individual the variant was observed to be de novo. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. For these reasons, this variant has been classified as Pathogenic.
