NM_152906.7(TANGO2):c.95G>A (p.Arg32Gln) was classified as Uncertain significance for Recurrent metabolic encephalomyopathic crises-rhabdomyolysis-cardiac arrhythmia-intellectual disability syndrome by Broad Center for Mendelian Genomics, Broad Institute of MIT and Harvard, citing ACMG Guidelines, 2015: The p.Arg32Gln variant in TANGO2 has not been previously reported in individuals with metabolic encephalomyopathic crises, recurrent, with rhabdomyolysis, cardiac arrhythmias, and neurodegeneration (MECRCN) but has been identified in 0.005% (1/18376) of East Asian chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP ID: rs896249235). Although this variant has been seen in the general population in a heterozygous state, its frequency is low enough to be consistent with a recessive carrier frequency. This variant has also been reported in ClinVar (Variation ID#: 803644) and has been interpreted as likely pathogenic by Mendelics. Computational prediction tools and conservation analyses do not provide strong support for or against an impact to the protein.
